چکیده

Hydroperoxide lyases cleave hydroperoxide of linoleic and linolenic acid to form C6-aldehyde and/or C9-aldehyde. These enzymes are widespread in plants [1] but there exist various types of substrate specificity. HPO lyase solubilized from tea chloroplasts can act on 13-HPO but not on 9-HPO [2] and vice versa in the case of the enzyme partially purified from pear fruits [3], Extracts of Phaseolus vulgaris leaves can cleave both 9-HPO and 13-HPO [4], To elucidate the physiological role of these enzymes and products, i.e. C6and C9-aldehydes, we have been searching for suitable enzyme sources. In this way we found that extracts of cucumber seedlings could cleave both 9and 13-HPO at various efficiencies peculiar to each organ.

Effect of volatile methyl jasmonate on the oxylipin pathway in tobacco, cucumber, and arabidopsis.

The effect of atmospheric methyl jasmonate on the oxylipin pathway was investigated in leaves of tobacco (Nicotiana tabacum L.), cucumber (Cucumis sativa L.), and Arabidopsis thaliana (L.). Differential sensitivities of test plants to methyl jasmonate were observed. Thus, different concentrations of methyl jasmonate were required for induction of changes in the oxylipin pathway. Arabidopsis was...

Lipoxygenase and hydroperoxide lyase in germinating watermelon seedlings.

Lipoxygenase (EC 1.13.1.13) was found in seedlings of Citrullus lanatus (Thunb.) Matsum. and Nakai (watermelon). The enzyme has pH optima of 4.4 and 5.5 and is inhibited by 0.2 mM nordihydroguaiaretic acid. It is present in two functional units with estimated molecular weights of 120,000 and 240,000, respectively.A new enzyme, tentatively termed hydroperoxide lyase, has been partially purified ...

Alfalfa contains substantial 9-hydroperoxide lyase activity and a 3Z:2E-enal isomerase.

Fatty acid hydroperoxides formed by lipoxygenase can be cleaved by hydroperoxide lyase resulting in the formation of short-chain aldehydes and omega-oxo acids. Plant hydroperoxide lyases use 13- or 9-hydroperoxy linoleic and linolenic acid as substrates. Alfalfa (Medicago sativa L.) has been reported to contain a hydroperoxide lyase specific for 13-hydroperoxy linoleic and linolenic acid only. ...

Profiling of Volatile Compounds and Associated Gene Expression and Enzyme Activity during Fruit Development in Two Cucumber Cultivars

Changes in volatile content, as well as associated gene expression and enzyme activity in developing cucumber fruits were investigated in two Cucumis sativus L. lines (No. 26 and No. 14) that differ significantly in fruit flavor. Total volatile, six-carbon (C6) aldehyde, linolenic and linoleic acid content were higher during the early stages, whereas the nine-carbon (C9) aldehyde content was hi...

Isolation, Expression, and Characterization of a Hydroperoxide Lyase Gene from Cucumber

A full-length cDNA coding for hydroperoxide lyase (CsHPL) was isolated from cucumber fruits of No. 26 (Southern China type) and No.14-1 (Northern China type), which differed significantly in fruit flavor. The deduced amino acid sequences of CsHPL from both lines show the same and significant similarity to known plant HPLs and contain typical conserved domains of HPLs. The recombinant CsHPL was ...
